La demencia tipo Alzheimer implica un deterioro severo en las funciones cognitivas
y ejecutiva y representa un 60-70% del total de los casos. A nivel neuropatológico,
se caracteriza por el depósito de péptido β-amiloide en placas neuríticas
extracelulares y la formación de ovillos neurofibrilares intraneuronales, cuya
eliminación ha sido el infructuoso objetivo de diferentes terapias. Por el contrario,
poco se sabe de la neuroinflamación existente, caracterizada por el aumento
del número y cambios morfológicos de las células microgliales en estadios avanzados
de la enfermedad.
El objetivo de este estudio es caracterizar las diferencias morfológicas de las
células de microglía en el hipocampo de un modelo murino añoso de enfermedad
de Alzheimer con el fin de dilucidar si, asociado con la edad, existe un fenotipo
activo proinflamatorio diferente al patrón fisiológico.
Se utilizaron 12 ratones hembra (dos grupos, n = 6 por grupo) de entre 19 y 22
meses. El grupo control fueron animales sanos y, el experimental, animales 3xTgAD.
Se seleccionaron aleatoriamente 10 células de cada animal y se usaron
las extensiones AnalyzeSkeleton y FracLac del programa Image-J para analizar
los distintos parámetros de las células. La ausencia de diferencias significativas
nos dice que debemos tener en cuenta la variable edad para entender el funcionamiento
de estos grupos celulares en el proceso de neurodegeneración, y que
debemos seguir estudiándolos, porque comprender su heterogeneidad morfológica
puede ser una de las claves. [-]

Alzheimer’s dementia involves severe impairment of cognitive and executive
functions and represents 60-70% of all cases. At the neuropathological level, it is characterized by the deposition of β-amyloid peptide in extracellular neurotic
plaques and the formation of intraneuronal neurofibrillary tangles, whose elimination
has been the unsuccessful goal of different therapies. In contrast, little is
known about the existing neuroinflammation, characterized by an increase in the
number and morphological changes of microglial cells in advanced stages of the
disease.
The aim of this study is to characterize the morphological differences of microglial
cells in the hippocampus of an aged murine model of Alzheimer’s disease
to elucidate whether, associated with age, there is an active proinflammatory
phenotype different from the physiological pattern.
Twelve female mice (two groups, n = 6 per group) aged between 19 and 22
months were used. The control group was healthy animals, and the experimental
group was 3xTgAD animals. Ten cells were randomly selected from each animal
and the AnalyzeSkeleton and FracLac extensions of the Image-J program were
used to analyze the different cell parameters. The absence of significant differences
tells us that we must consider the age variable to understand the functioning
of these cell groups in the neurodegeneration process, and that we must
continue to study them, because understanding their morphological heterogeneity
may be one of the keys of this neurodegenerative disorder. [-]
