dc.description.abstract | Alzheimer's disease (AD) is a progressive and irreversible neurodegenerative disorder of the central nervous system recognized as "Major Neurocognitive Disorder" in the DSM-V. This disorder primarily affects people over 65 years old and is the most frequent current cause of dementia in the world, representing 60-70% of all cases. Regrettably, it’s etiology is unknown nowadays, as well as an effective treatment for it. In order to have an AD model that reproduces the neuronal pathology and the typical symptomatology of this disease, a large number of transgenic mice have been characterized. In this work, we have used male transgenic triple mutant mice (3xTg-AD) for Alzheimer's disease (APPswe, PS1M146V and Tau P301L), as well as control mice with wild type B6129SF2/J. As a reflection of what is observed in humans with AD, the 3xTg-AD mice studied presented cognitive and behavioral deficits, as well as motor deficits and β- amyloid accumulations (Aβ) and tau in their brains. The main objective of this study was to determine if there were cognitive, behavioral and other kind of alterations observed in human with AD as anxiety in the triple transgenic model of AD, and to verify if these alterations corresponded with histological alterations in the brain of the murines studied in their different ages (3 and 6 months). With this purpose, several behavioral tests (Elevated plus maze and Open field test) were carried out. Also a postmortem histological test was made, observing a higher level of anxiety and hypomobility in the male transgenic mice. dc.description.abstract | La enfermedad de Alzheimer (EA) es un desorden neurodegenerativo progresivo e irreversible del sistema nervioso central reconocido como “Desorden Neurocognitivo Mayor” en el DSM-V, que afecta fundamentalmente a personas mayores de 65 años y es la causa actual más frecuente de demencia en el mundo, representando el 60-70% de todos los casos, y de la que lamentablemente, a día de hoy, se desconoce la etiología así como un tratamiento eficaz. Con el objetivo de disponer de un modelo de EA, que reproduzca la patología neuronal y la clínica típica de esta enfermedad, se han caracterizado una gran cantidad de ratones transgénicos. En este trabajo hemos empleado ratones hembras mutantes triple transgénico (3xTg-AD) para la enfermedad de Alzheimer (APPswe, PS1M146V y Tau P301L), así como ratones control con genotipo salvaje B6129SF2/J. Como reflejo de lo que se observa en humanos con EA, los ratones 3xTg-AD estudiado presenta déficits cognitivos y conductuales, así como déficits motores y acumulaciones β-amiloide (Aβ) y tau en sus cerebros. El principal objetivo planteado con este estudio fue dilucidar si en el modelo triple transgénico de EA existían alteraciones cognitivas, conductuales y demás alteraciones observadas en la EA humana como ansiedad, y comprobar si éstas alteraciones se correspondían con alteraciones histológicas en el cerebro de los murinos estudiados en diferentes edades (3 y 6 meses). Con éste fin, se llevaron a cabo diversas pruebas conductuales (prueba del laberinto elevado y prueba de campo abierto), y asimismo una comprobación histológica postmortem, observando un mayor nivel de ansiedad e hipomovilidad en los ratones transgénicos hembras, y que se reflejó en un mayor área marcada en ciertos núcleos con cFos. | ca_CA |
