Introducción: el glioblastoma multiforme es el tumor primario cerebral más frecuente y agresivo, y
se encuentra asociado a una elevada mortalidad. La respuesta al tratamiento estándar es insuficiente
en la mayoría de los casos, siendo frecuente la recaída. Por tanto, el desarrollo de nuevos
tratamientos más efectivos en base a las alteraciones subyacentes del tumor es de suma
importancia. Objetivos: la finalidad de este estudio es comprobar la reproducción de las
características del GBM humano mediante modelos PDOX en ratones. De manera secundaria,
mostrar qué nuevas alternativas terapéuticas han tenido éxito en estos modelos preclínicos.
Metodología: esta revisión incluye un total de 18 estudios preclínicos extraídos a partir de una
búsqueda estructurada en las bases de datos PubMed y Cochrane Library. Los datos relevantes de
cada estudio se sintetizaron en una tabla para su posterior análisis cualitativo (crecimiento y tamaño
tumoral, integridad de la barrera hematoencefálica, etc.) o cuantitativo (supervivencia media de los
sujetos en función del tratamiento). La calidad metodológica de cada publicación también fue
analizada. Resultados: en cuanto a las características básicas del tumor (crecimiento tumoral, etc.),
los modelos PDOX reproducen el GBM humano. Entre los 18 estudios incluidos, 15 descubrieron
fármacos con eficacia terapéutica, en base a las diferentes medidas de resultado analizadas. De ellos,
13 aumentaron de manera significativa la supervivencia media alcanzada. Conclusión: los modelos
PDOX reproducen las principales características del GBM humano. La mayoría de fármacos probados
han demostrado eficacia terapéutica. Estos resultados demuestran la utilidad de los PDOX, y animan
a continuar investigando en esta línea. [-]

Introduction: glioblastoma multiforme is the most frequent and aggressive primary brain tumour,
and it is associated with high mortality. The response to standard treatment is insufficient in most of
cases, with relapse being frequent. Therefore, the development of new more effective treatments
based on the underlying alterations of the tumour is absolutely crucial. Objectives: the main aim of
this study is to verify that PDOX models can recapitulate the characteristics of human GBM in mice.
Secondarily, to show the new therapeutic alternatives that have been successful in these preclinical
models. Methodology: this review includes 18 preclinical studies extracted from a structured
research in PubMed and Cochrane Library databases. The relevant data from each study were
synthesized in a table for further qualitative (tumour growth and size, integrity of the blood-brain
barrier, etc.) or quantitative analysis (average survival of subjects according to treatment). The
methodological quality of each publication was also analyzed. Results: regarding the basic5
characteristics of the tumour (tumour growth, etc.) the PDOX models recapitulate the human GBM.
Among the 18 included studies, 15 discovered drugs with therapeutic efficacy, based on the different
outcome measures analyzed. Of these, 13 significantly increased the average survival achieved.
Conclusion: PDOX models recapitulate the main characteristics of human GBM. Most drugs tested
have proved therapeutic efficacy. These results demonstrate the utility of PDOX, and encourage
further research along these lines. [-]
