dc.description.abstract | Introduction : V ariation in pigmenta ry traits is a striking example of human phenotypic diversity. Genetic va riant s involved in the pigmentation path way are associated with varia bility in the pigmentation of the iris, especially in blue and brown eyes. However, little is known about the genetic determinants that define green - coloured iris and the peripu p il lary ri ng. Objectives : To a nalyse the genetic variations that define the patterns of iris pigmentation and to evaluate the clinical usefulness of the study of these genes. Methodology : DNA samples from 829 volunteers were obtained to determine the genotype of 8 g enes involved in pigmentation. Subsequently , the association between eye colour and genotype was analysed using logistic regression. Results : We confirmed that the SNP rs12913832 in HERC2/OCA2 is the major eye colour determinant. Although the derived alle le C is associated with blue eye colour, many individuals with green eyes are heterozygous carriers. Polymorphisms in IRF4 and OCA2 also seem to have an influence on green iris es . An association between the presence of peripupillary ring and the heterozygo us state in SNP rs12896399 in the SLC24A4 gene has also been observed. Conclusion : Eye colour prediction tools should take into account inheritance models , since the number of copies of the derived allele influences iris pigmentation. | ca_CA |
dc.description.abstract | Introducción : L a variación de los rasgos de pigmentación es un ejemplo llamativo de la diversidad fenotípica humana . Variantes genéticas involucrad a s en la ruta de la pigmentación se asocian claramente a la variabilidad en la pigmentación del iris, especialmente en ojos azules y marrones. Sin embargo, se conoce poc o de los determinantes genéticos que definen el iris de color verde y el anillo peripupilar. Objetivos : Analizar las variaciones genéticas que definen los patrones de pigmentación del iris y evaluar la utilidad clínica del estudio de dichos genes . Metodolo gía : Obtuvimos muestras de ADN de 829 voluntarios para determinar el genotipo de 8 genes involucrados en pigmentación. Analizamos la a sociación entre color de ojos y genotipo mediante regresión logística. Resultados : Se confirma que el SNP rs12913832 en HE RC2/OCA2 es el mayor determinante en el color de ojos. Aunque el alelo derivado C se asocia a ojos azul es , muchos individuos con ojos verdes son portadores heterocigotos . Polimorfismos en IRF4 y OCA2 también parecen influir en el iris verde. O bservamos una asociación entre anillo peripupilar y ser heterocigoto para rs12896399 en SLC24A4 . Conclusión : Las herramientas de predicción del color de ojos deberían tener en cuenta modelos de herencia, ya que el número de copias del alelo derivado influye en la pigme ntación del iris. | ca_CA |
