Las patologías que afectan al epitelio pigmentario de la retina provocan una
importante pérdida de función visual, siendo el estrés oxidativo, una causa de
disfuncionalidad del tejido. En la actualidad, las posibilidades terapéuticas para su
manejo son escasas. Dentro del avance en el campo de la medicina regenerativa, las
células madre mesenquimales, como las del cuello uterino humano (hUCESCs), podrían
contribuir al mantenimiento de la homeostasis de la retina. Así, hemos investigado el
efecto del medio condicionado de las hUCESCs (MC-hUCESCs) sobre una línea
comercial de epitelio pigmentario de la retina (ARPE-19), en condiciones de estrés
oxidativo. Por otro lado, estudiamos los efectos in vitro del MC-hUCESCs en la
formación de tubos en células endoteliales de vena umbilical humana (HUVEC). Tras
incubar las ARPE-19 con H₂O₂ en presencia o no de MC-hUCESCs, se analizó mediante
qPCR, la expresión de factores como HO-1, gen antioxidante, y VEGFA, PDGFA y
PDGFB, relacionados con la vasculogénesis. La expresión de ARNm de HO-1, VEGFA,
PDGFA y PDGFB aumentó de forma significativa cuando las células ARPE-19 fueron
tratadas con H₂O₂ en presencia de MC-hUCESCs con respecto a las células tratadas
únicamente con H₂O₂. En cuanto al ensayo de formación de tubos, las HUVEC tratadas
con sobrenadante de células ARPE-19 tratadas con H₂O₂ + MC-hUCESCs mostraron un
aumento significativo en la longitud media de los vasos, el número de uniones
capilares y el área media de los vasos, en comparación con las HUVEC tratadas con
sobrenadante de células ARPE-19 tratadas solo con H₂O₂. Los resultados del presente
estudio muestran el potencial terapéutico del MC-hUCESCs sobre el epitelio
pigmentario de la retina ante el daño ocasionado por el estrés oxidativo, como es la
estimulación de un factor antioxidante y una mejoría de la vascularización. [-]

The pathologies that affect the retinal pigment epithelium cause a significant loss of
visual function, with oxidative stress being a cause of tissue dysfunction. At present,
the therapeutic possibilities for its management are scarce. Within the advance in the
field of regenerative medicine, mesenchymal stem cells, such as human uterine
cervical stem cells (hUCESCs), could contribute to the maintenance of the retinal
homeostasis. So, we have investigated the effect of the conditioned medium of
hUCESCs (MC-hUCESCs) on a commercial line of retinal pigment epithelium (ARPE-19),
under conditions of oxidative stress. On the other hand, we studied the in vitro effects
of MC-hUCESCs on the formation of tubes in human umbilical vein endothelial cells
(HUVEC). After the ARPE-19 were incubated with H₂O₂ in the presence or absence of
MC-hUCESCs, the expression of factors such as HO-1, antioxidant gene and VEGFA,
PDGFA and PDGFB, related to vasculogenesis, was analyzed by qPCR. The mRNA
expression of HO-1, VEGFA, PDGFA and PDGFB increased significantly when ARPE-19
cells were treated with H₂O₂ in the presence of MC-hUCESCs relative to cells treated
only with H₂O₂. Regarding the tube formation test, HUVEC treated with supernatant
from ARPE-19 cells treated with H₂O₂ + MC-hUCESCs showed a significant increase in
average vessels length, number of capillary-like junctions and average of vessel,
compared with HUVEC treated with supernatant from ARPE-19 cells treated with H₂O₂
only. The results of the present study show the therapeutic potential of MC-hUCESCs
on the retinal pigment epithelium in the face of damage caused by oxidative stress,
such as the stimulation of an antioxidant factor and an improvement in vascularization. [-]
