Spinal muscular atrophy (SMA) is the hereditary disease that causes the most deaths in childhood. Although there is no cure , gene therapy offers the most promising alternatives in the development of their treatment, with antisense oligonucleotide therapy being the only approved treatment that modifies the evolution of the disease.
This review’s aim is to compare the efficacy and safety profile of
gene therapy treatments for AME. To do this , 3 clinical trials were analyzed on two different therapies ; selected after a structured Pubmed search. The variables of survival, motor function and adverse events were studied. . To evaluate the methodological quality, the Cochrane Handbook 5. 1. 0 part 2 w as used. Both therapies have demonstrated, taking into account the limitation of their current clinical trial’s phase, to improve the survival and motor function of patients with SMA. The heterogeneity between the samples of both therapies prevents a well-founded comparison. More over, one of them is a Phase I clinical trial, thus the results should not be extrapolated. Therapy with ASOs is the best therapeutic treatment at present. [-]

La atrofia muscular espinal (AME) es la enfermedad hereditaria que más muertes causa en la infancia. Aunque no existe cura, la terapia génica ofrece las alternativas más prometedoras en el desarrollo de su tratamiento, siendo la terapia con oligonucleótidos antisentido el único tratamiento aprobado que modifica la evolución de la enfermedad. Esta revisión pretende comparar el perfil de seguridad y e ficacia de los tratamientos para la AME basados en la terapia génica. Para ello, se analizan 3 ensayos clínicos sobre dos terapias distintas, seleccionados mediante una búsqueda estructurada en Pubmed. Se estudiaron principalmente las variables de supervivencia, función motora y eventos adversos . Para evaluar la calidad metodológica se empleó el Manual de Cochrane 5. 1. 0 parte 2. Ambas terapias han demostrado, teniendo en cuenta la limitación de la fase del ensayo en la que se encuentran, mejorar la supervivencia y la función motora de los enfermos con AME. La heterogeneidad entre las muestras de ambas terapias impide una comparación fundamentada y siendo una de ellas Fase 1, no deben extrapolarse los resultados. La terapia con ASOs es la mejor opción terapéutica en la actualidad. [-]
