Relaxin-3(RLN3) es un miembro de la superfamilia de la insulina. Su principal lugar de expresión es el nucleus incertus del tronco encefálico, desde donde las neuronas envían proyecciones ascendentes el mesencéfalo, hipotálamo y telencéfalo. Este neuropéptido presenta una alta afinidad por el receptor acoplado a proteína G RXFP3. Dicho receptor expresa ampliamente en diversas zonas del SNC entre las que se encuentran las zonas de proyección del nucleus incertus. Estudios previos han sugerido un importante papel de RLN3 como modulador de respuestas de estrés y control de apetito, entre otras. El objetivo de este estudio era determinar qué núcleos hipotalámicos presentan abundancia de fibras de RLN3, y poder relacionar dichos hallazgos con el posible papel de RLN3 en la modulación de las conductas alimentarias y otras funciones metabólicas. Se usaron ratas macho Wistar y un protocolo de inmunohistoquímica de marcaje inmunocitoquímico para el estudio de la distribución de RLN3 en cuatro niveles hipotalámicos. Detectamos abundancia de fibras en hipotálamo anterior (AH), lateral (LH) y posterior (PH), tres de los núcleos que se conocen implicados en el control del apetito y la saciedad, así como en núcleo dorsomedial hipotalámico, núcleo preóptico lateral LPO, núcleo paraventricular anterior y núcleo supramamilar. Estos resultados refuerzan la hipótesis de la implicación de RLN3 en el control de la ingesta. [-]

Relaxin-3 (RLN3) is a neuropeptide of the insulin superfamily. It is mainly expressed in the nucleus incertus of the brainstem, from where the neurons send ascending projections towards the midbrain, hypothalamus and telencephalon. This neuropeptide displays high affinity to a G-protein coupled receptor RXFP3. This receptor is widely distributed in areas projected by the nucleus incertus. Previous studies have suggested an important role of RLN3 as a modulator of stress responses and food intake control, among others. The aim of this study was to determine which hypothalamic nuclei are targeted by RLN3fibers, and to relate these findings to the possible role of RLN3 in the modulation of feeding behaviors. An inmunohistochemistry protocol was used in male Wistar rats to study the distribution of RLN3 at four hypothalamic levels. We detected abundance of fibers in the anterior hypothalamus (AH), lateral (LH) and posterior (PH), three of the nuclei known to be involved in appetite control and satiety, as well as dorsomedial hypothalamic nucleus, lateral preoptic nucleus (LPO), anterior paraventricular nucleus and supramammillary nucleus.
