In recent decades, pharmaceutical industry has been investigating novel strategies
to develop pharmaceuticals and drugs. Nowadays, nearly 700 new drugs derives from
natural products, which are chemical compounds or substances that could be found in
nature. They could be prepared by chemical synthesis (semi- and total synthesis), playing
an important role in the development of organic chemistry, due to their wide functional
diversity.
In this work, the example of ingenol mebutate, a hydrophobic macrocyclic diterpene
ester isolated from Euphorbia peplus, was used, performing a review of different synthesis
of this compound. This compound is a natural product, which was later approved as a
therapeutic agent for actinic keratosis, sun-induced lesion. It shows elicit activity through
a dual mechanism of action: recruitment of innate immune system and induction of necrosis
that specifically targets dysplastic cells, as well as neutrophil-mediated immunostimulatory
effects.
Three different synthesis of ingenol mebutate skeletal core and three total synthesis
of this compound were reviewed, highlighting the differences between them. [-]
