La enfermedad de Alzheimer (EA) es la causa principal de demencia entre la población
mundial. Se trata de una enfermedad neurodegenerativa, de etiología desconocida hasta la
fecha, aunque durante décadas se han propuesto diversas hipótesis sobre su origen. La
principal es la que relaciona la EA con el acúmulo cerebral de beta-amiloide (Aβ), siendo
este el responsable en última instancia de la alteración a nivel de las sinapsis neuronales.
Por otra parte también se ha propuesto la hipótesis relacionada con la proteína tau y su
hiperfosforilación, lo cual acaba derivando en el acumulo de ovillos neurofibrilares. Ambos
procesos inducirían ulteriormente una pérdida neuronal asociada con el deterioro cognitivo.
Además, recientemente se han establecido relaciones entre el acúmulo proteico
cerebral y los estados de neuroinflamación, los cuales están mediados por diversos
componentes celulares, entre ellos las células microgliales. También se han encontrado
analogías entre estos estados inflamatorios, y los derivados de los estados hipoglucémicos,
producido entre otras patologías por la Diabetes Mellitus tipo 2.
En este trabajo se ha buscado establecer la relación entre los acúmulos cerebrales de
Aβ y el aumento de microglía cerebral, tanto en el septum medial como en la banda
diagonal, indicativas de neuroinflamación. Además se ha buscado estudiar las proyecciones
existentes de relaxina tipo 3 (RLN3), péptido perteneciente a las insulinas, desde estas
zonas cerebrales.Alzheimer’s disease (AD) is the main cause of dementia among the world’s population. It
is a neurodegenerative disease, but nowadays its etiology is unknown, and during decades
there have been proposed various hypotheses about its origin. The principal one is the
hypothesis that relates the AD with the cerebral accumulation of beta-amyloid protein (Aβ),
being this the latest responsible of the synaptic alteration. It has also been proposed another
hypothesis related to the tau protein and its hyperphosphorylation, deriving from this process
the accumulation of neurofibrillary tangles. Both processes would subsequently induce a
neuronal loss, which will be associated with the cognitive impairment.
Besides, recently has been established a relation between the protein accumulation in
the brain and neuroinflammation states. Those states are mediated by various cellular
components, including the microglial cells. Analogies have been also found between those
neuroinflammatory states and the ones derived from the hyperglycemia, which are produced
among other pathologies, by the Diabetes Mellitus Type 2 (DMT2).
In this study, we have sought to establish a relation between the cerebral accumulation
of Aβ and the microglia increase, both in medial septum and in diagonal band. We have also
sought to study the relaxin-3 (RLN3) projections from this cerebral areas, being this a peptide
belonging to the insulin family.
